PDRN Polynucleotide Therapy: The Newer Skin Regenerator, Explained

PDRN — polydeoxyribonucleotide — is one of the more interesting newer entries in regenerative aesthetic medicine. It's distinct from PRP, distinct from HA fillers, and distinct from biostimulators like Sculptra. Whether it belongs in your treatment plan depends on what you're trying to do.
What PDRN actually is
PDRN is a fragment of DNA — specifically, polynucleotide chains derived from purified salmon trout DNA. The molecule is structurally similar enough to human DNA that it interacts with adenosine receptors and trophic pathways in human cells. Functionally, it acts as a regenerative signal: stimulating fibroblasts to produce more collagen and elastin, accelerating tissue repair, and reducing local inflammation.
It's not filler — there's no real "volumizing" effect. It's a signaling molecule that tells your tissue to behave more like younger, healthier tissue.
How it differs from PRP and from fillers
- vs. PRP: PRP delivers your own concentrated platelets and growth factors. PDRN delivers a synthesized regenerative molecule from salmon DNA. Both stimulate fibroblasts; PDRN does so more consistently across patients (PRP quality varies with the donor's health and platelet count). PDRN doesn't require a blood draw.
- vs. HA fillers: Fillers add volume mechanically. PDRN doesn't add volume — it improves the quality of the tissue you have. The effect is subtle and cumulative, not immediate.
- vs. Sculptra: Sculptra is a collagen biostimulator that works over 4-6 months as the body builds collagen around PLLA particles. PDRN works on a faster timeline (visible improvement at 3-4 weeks) but produces less dramatic structural change. They can be paired.
What it actually does
The clinical applications with the strongest evidence base:
- Under-eye rejuvenation: One of PDRN's signature uses. Improves dark circles caused by thin/poor-quality tissue, reduces fine lines, and can soften discoloration. Series of 3-4 treatments spaced 2-3 weeks apart.
- Skin texture and tone: Across face, neck, décolleté. Best for skin that's structurally sound but looks dull or lacks "bounce." Series of 3-4 sessions.
- Scar improvement: Including atrophic acne scars and post-surgical scars. Often paired with microneedling or laser.
- Hair restoration: Similar to PRP — extends follicle activity in early-stage thinning. Some practices prefer PDRN over PRP for hair work.
- Wound healing: Used internationally for slow-healing wounds, post-surgical recovery.
Who's a good candidate
- Clients in their 30s-50s who want gradual improvement in skin quality without filler or downtime.
- Anyone with mild-to-moderate under-eye concerns where filler would be too aggressive.
- Clients with acne scarring looking for an additional regenerative input alongside microneedling.
- Anyone wanting a "preventive" treatment that maintains tissue quality.
Who's not
- Anyone allergic to fish or fish DNA (the source material).
- Active autoimmune flares.
- Active cancer or pregnancy.
- Clients hoping for visible filler-style change in one session.
The procedure
PDRN is injected as a series of small intradermal or subcutaneous deposits across the treatment area. Topical numbing is applied; the injections themselves are well-tolerated. A facial session typically takes 30-45 minutes total.
Mild redness, very slight bumps at injection points, and brief skin flushing are normal for 24-48 hours. Most clients return to normal activities the same day.
Results timeline
- Days 1-3: Mild local response resolves.
- Weeks 2-4: Initial improvement in skin quality, hydration, and softness becomes noticeable.
- Months 1-3: Cumulative improvement across the series. Each session builds on the last.
- Month 6: Maintenance can begin. Most clients do a single touch-up every 6-12 months.
Combining with other treatments
PDRN pairs well with:
- Microneedling — applied topically through microchannels for synergistic effect.
- Sculptra — Sculptra builds the structural collagen, PDRN refines the surface tissue quality.
- Botox — works on different layers (PDRN on tissue quality, Botox on muscle movement).
- HA fillers — fillers volumize, PDRN improves the surrounding tissue around the filler.
